چکیده

A recently developed procedure for alkylatiori of organic acids has proven extremely successful for preparation of volatile alkyl derivatives of the barbiturates for gas-chromatographic analysis. Soluble salts are formed in a mixture of N,N-dimethylacetamide and methanol. These in turn react rapidly with alkyl iodides to form the corresponding alkyl derivatives. The butyl derivatives of barbiturates, prepared in this manner, are separable by high-resolution gaschromatography. Any of 14 barbiturates can be determined simultaneously or separately (although there is some overlap with certain uncommon barbiturates). The butyl derivatives of several barbiturates that are unresolved in the form of underived compounds or methyl esters can be resolved, thus overcoming many previous analytical limitations.

An Improved GC Method for Rapid Analysis of Valproic Acid in Human Plasma Without Derivatization

A simple, reproducible and rapid gas chromatographic method for precise determination of valproic acid (VPA) in human plasma has been developed. Total time for sample preparation and GC analysis is less than 45 min. After plasma protein precipitation, VPA was extracted into chloroform with suitable recovery. By using Stabilwax®<span styl...

A Sensitive Liquid Chromatographic Method for the Analysis of Clarithromycin with Pre-Column Derivatization: Application to a Bioequivalence Study

Objective(s) A sensitive liquid chromatographic method for the analysis of clarithromycin- a macrolide antibiotic- in human serum, using pre-column derivatization with 9-fluorenylmethyl chloroformate (FMOC-Cl) is described. Materials and Methods The method involved liquid-liquid extraction of the drug and an internal standard (amantadine) followed by pre-column derivatization of the analytes...

A gas-chromatographic method for separating and measuring barbiturates and glutethimide in blood.

A gas-chromatographic method is described for the quantitative deter. mination of 11 barbiturates and glutethimide in blood. The extracted barbiturates are methylated and injected into a 7% DC-200 column. Use of methohexital as the internal standard, as well as the addition of standards to a drug-free sample of blood, compensates for losses and for differences in the distribution coefficient of...

Enantioselective Gas Chromatographic Separation of Racemic N-alkylated Barbiturates: Application of C11-Chirasil-Dex as Chiral Stationary Phase in GC

Chirasil-beta-Dex containing an undecamethylene spacer (C11-Chirasil-Dex) was synthesized and used as chiral stationary phase (CSP) in enantioselective gas chromatography (GC). The versatility of the new stationary phase in the simultaneous enantiomeric separation of a set of N-alkylated barbiturates is demonstrated.

Derivatization Reactions and Reagents for Gas Chromatography Analysis

Derivatization reactions are meant to transform an analyte for detectability in Gas Chromatography (GC) or other instrumental analytical methods. Derivatization in GC analysis can be defined as a procedural technique that primarily modifies an analyte’s functionality in order to enable chromatographic separations. A modified analyte in this case will be the product, which is known as the deriva...
